Utilization of plant bioindicators in monitoring of environmental contamination

Environmental contamination may be observed by different analytical tools as well as by various types of bioindicators providing relevant information about the effects of toxic substances and their mixtures towards organisms in situ. The aim of this work is to provide a comprehensive overview of plant bioindicators and biomonitors suitable for biomonitoring of persistent organic pollutants (POPs) in natural ecosystems. For groups of organic pollutants such as PCBs, PCDDs / Fs, PBDEs, PAHs, PFCs, and OCPs, review of literature is provided including knowledge about different species of coniferous trees, angiospermous plants (including crops), mosses and lichens that can be used for monitoring. In this work, analysis examples of samples of pine needles, bark, moss and lichen tissue are quoted as biomonitor tools. An overview of plants for phytotoxicity and genotoxicity observation using pollen grains abortivity and online bioindication as an example on non-specific biomonitoring is also provided.
Trends in tree-ring width of treeline trees along latitudinal gradient from subtropics to the Arctic.

This bachelor thesis is interested in a tree ring of trees at the treeline from subtropics to the arctic and the main aim of this work is to find if there is a trend in the tree ring width at the treeline along the latidunial gradient. The work/thesis focuses on conifers. The data were obtained from International Tree-Ring Data Bank database, article searches and from the internal workplace database. A total of 9859 annual ring samples were obtained from 153 localities in Europe. Trees of the genus Pine (Pinus), Spruce (Picea) and Fir (Abies) were selected, which dominate the europeans ecotones of treeline. After removing the age trend, the regression of tree ring width and the latitude, were calculated. The results showed that in most cases the tree ring width decreases with higher latitude. However, a small number of tree ring samples from latitudes around 55ř north latitude adds to the uncertainty of this claim.
Silicified stems of upper Paleozoic plants from the Intra Sudetic and Krkonoše Piedmont basins

The late Paleozoic deposits of the Czech Republic are famous for their rich occurence of silicified stems. Despite the fact they have been often described and are well-known among scientists and collectors, their modern evaluation is lacking. This work summarizes results of recent anatomical and paleoenvironmental studies of silicified stems of the Intra Sudetic and Krkonoše Piedmont basins, where are these fossils found very frequently. Based on field research and review of public and private collections, the presence of silicified remnants was proved in several stratigraphic units. Firstly, this work deals with silicified stems of calamitaleans, which are known from the Ploužnice Horizon of the Krkonoše Piedmont Basin, and some gymnosperms. Based on anatomical studies of the secondary xylem and other related features there were found two species of calamitaleans: Arthropitys cf. bistriata and Calamitea striata. Secondly, the more abundant Agathoxylon - type of wood was divided into two groups, which are assigned to cordaitaleans, and conifers. The palaeoenvironmental conditions were partly reconstructed according to sedimentary structures and also according to cordaitaleans - conifers ratio in each wood-bearing layer. Analysis of primary photosynthetic processes in conifers: A comparison of selected methods and their possible utilisation for the study of genetic variability

Conifers are important both ecologically and socioeconomically, however, same parts of their biology are not that well researched. This includes genetics and breeding and partly even physiology. Because quantitative genetic analyzes applied in breeding necessitate an analysis of a large number of samples, and conventional methods of analysis are quite time-consuming, certain parameters describing e.g. the activity of photosynthetic electron-transport chain (ETC) are considered for such use. Several methods of the measurement of the activity of photosynthetic ETC exist, but there are some problems with their usage in conifers. I studied this issue from different points of view in three parts of this thesis. 1) I compared the photosynthetic ETC activity in 8 species of conifers using chlorophyll (Chl) fluorescence measurements on intact needles and polarographic measurements in isolated chloroplasts. Each method brought different information. 2) I measured Chl fluorescence parameters, reflectance spectra and pigment content in 536 genetically defined trees of Pinus sylvestris L. Changes in anatomy of coniferous needles in response to anthropogenic pollution

The anatomical structure of conifer needles is affected by external enviromental conditions. From abiotic factors, apart of irradiation, main determining factors are drought and extreme temperatures belonging to factors of climate change. Anthropogenic pollution has a significant position among the external abiotic factors that cause changes in the structure of needles. At present, the main polluting compounds are tropospheric ozone, SO2, NOx and PM. Due to the effect of atmospheric pollutants, the proportion of individual tissues in the needle changes, so does the proportion of intercellular spaces and the internal structure of cellular organelles, especially chloroplasts. Mesophyll cells show signs of degradation, especially around the substomatal cavity. There appears a visible damage on the needle surface - chloroses and necroses. The process of senescence speeds up during the activity of stress enviromental factors, such as anthropogenic pollution. The main objective of the present Thesis is to gather knowledge about changes of conifer needle structure, caused by anthropogenic pollution, especially ozone, SO2 and NOx, in combination with climate change factors - drought and extreme temperatures. Conifers in the education of the biology

3 Abstract This thesis covers conifers (Pinopsida) and their use in teaching natural science in primary schools and biology in secondary schools. The aim is to analyze the problems of conifers (Pinopsida) at FEP (Framework Education Programme) for basic education and in selected textbooks, to determine the level of teaching conifers (Pinopsida) in primary schools and secondary schools in the Benešov region and propose a project with the theme of conifers (Pinopsida) for primary and secondary schools, with partial verification in the classroom. The theoretical part is concerned with conifers (Pinopsida) from a biological point of view. Characterize a group of conifers (Pinopsida), it's representatives in the Czech Republic and also deal with the ecology of conifers (Pinopsida). In the research part, using text and tables I mention results of analysis of FEP for basic education and analysis of textbooks. Later in the form of tables and graphs I mention the results of research on teaching conifers (Pinopsida) in primary and secondary schools in the Benešov region. Research was conducted using the questionnaire. Finally, I present the proposed project: The scent of resin partially verified in primary school. This work is intended for all who are interested in the conifers (Pinopsida) from the didactic point of view.
